Phospholipid extraction methods

1.Folch Method:
Principle: Based on the original Folch extraction method, this technique involves a chloroform-methanol extraction.
Procedure:
Mix the sample with chloroform-methanol (2:1 ratio).
Add water to induce phase separation.
Centrifuge to separate phases.
Collect the lower organic phase (containing lipids) and evaporate the solvent.
2.Bligh and Dyer Method:
Principle: Similar to the Folch method, it uses a chloroform-methanol extraction but with a modified solvent ratio.
Procedure:
Mix the sample with chloroform-methanol (1:2:0.8 ratio).
Add water for phase separation.
Centrifuge to obtain lipid-containing chloroform phase.
Evaporate the solvent.
3.MTBE (Methyl Tert-Butyl Ether) Method:
Principle: Uses MTBE as a solvent, which is less toxic than chloroform.
Procedure:
Mix the sample with MTBE-methanol (3:1 ratio).
Add water for phase separation.
Centrifuge and collect the upper phase (organic phase).
Evaporate the solvent.
4.Hexane-Isopropanol Method:
Principle: Utilizes hexane and isopropanol for lipid extraction.
Procedure:
Mix the sample with hexane-isopropanol (3:2 ratio).
Centrifuge to obtain the organic phase.
Evaporate the solvent.
5.Saponification Method:
Principle: Involves saponifying lipids to release fatty acids and glycerol, followed by extraction of phospholipids.
Procedure:
Saponify lipids with alkali.
Acidify the solution to release phospholipids.
Extract phospholipids with an organic solvent.
6.Solid-Phase Extraction (SPE):
Principle: Uses a solid-phase matrix to selectively retain phospholipids while other components are washed away.
Procedure:
Sample is loaded onto an SPE cartridge.
Phospholipids are retained, and contaminants are washed away.
Elute the phospholipids with an appropriate solvent.
